Animal models are critical in the selection and development of novel therapeutics. Studies evaluating exposure, tolerability, dose-exposure linearity, bioavailability, and efficacy in a sentinel screening model are often used when selecting a lead clinical candidate. Further insight into the therapeutic potential of a compound can be gained through scheduling and combination studies and the exploration of additional disease models. The likelihood of clinical success can be increased through an early understanding of the mechanism of action, PK-PD relationship, metabolism, toxicology and the development of biomarkers and a translational path.
